PNC-27

PNC-27 is a synthetic 32-amino acid peptide derived from the p53 tumor suppressor protein. It was designed to selectively target and destroy cancer cells while leaving healthy, normal cells completely unharmed. It works by binding to HDM-2 proteins found on the surface of cancer cell membranes — a marker that normal cells do not express — making it one of the most targeted anticancer peptides currently being researched.

HOW IT WORKS

PNC-27 contains the HDM-2 binding domain of the p53 protein, linked to a cell-penetrating sequence. When it encounters a cancer cell, it binds to the HDM-2 protein expressed on that cell’s membrane. This triggers the formation of transmembrane pores — tiny holes that disrupt the cancer cell’s membrane integrity, causing osmotic shock, cellular depolarization, and rapid necrotic cell death. The entire process is independent of the cell’s p53 status, meaning it works even on cancers that have mutated or disabled their p53 pathway.
